Foetid juniper (Juniperus foetidissima) is an important species in the Cupressaceae family holding ecological and biological value. In Iran, J. foetidissima species is only present in Arasbaran biosphere reserve. In the present study, the genetic diversity and structure of eight populations of J. foetidissima in the Arasbaran forest were evaluated using 6 polymorphic SCoT markers to select diverse individuals for forming a seed orchard. A total of 118 polymorphic bands were produced using six SCoT primers, indicating a polymorphism rate of 100% and an average of 19.6 polymorphic bands per SCoT. The number of polymorphic bands varied from 13 to 29, with SCoT10 and SCoT11 amplifying the maximum and minimum number of bands, respectively. The average polymorphic information content (PIC) for the primers was 0.321, with SCoT41 having the highest PIC value of 0.378 and SCoT10 having the lowest value of 0.279. The Average values of genetic diversity parameters, including the number of alleles (Na), effective number of alleles (Ne), Shannon’s information index (I), and expected heterozygosity (He) were 1.23, 1.29, 0.287 and 0.184, respectively. Analysis of molecular variance (AMOVA) revealed that 87% of the observed genetic differentiation was distributed within populations, while 17% occurred among populations, suggesting that the genetic diversity within populations was more significant than that among populations. The results of the UPGMA method categorized the 48 individuals from the 8 populations into 5 groups. The findings of this work can provide strategies for the maintenance, breeding, and conservation of J. foetidissima. | ||
