| In order to produce haploid plants through ovary culture in cucumber (Cucumis sativus L.), a factorial experiment based on randomized design was conducted with three replicates under in vitro conditions. The ovaries of Sina, a hybrid variety of cucumber were harvested one week after flowering. They were cultured in the MS medium containing different concentrations of (0, 0.8 and 1.2 mg/L) TDZ and BAP, respectively. The best calli and the highest percentage of calli were obtained in the culture medium containing 0.8 mg/L TDZ and BAP. Then, 3 week-old explants were transferred to the differentiation medium; MS medium, supplemented with NAA, IBA, IAA, 2,4-D, Pic at 0.05 mg/L and 3 cytokinins including TDZ, BAP, Kin with 0.5, 1 and 1.5 mg/L, respectively, with a total of 15 treatments and 3 replicates. They were subcultured once every two weeks. The highest percentage of callus formation was obtained in the presence of 0.05 mg/L NAA and 1.5 mg/L TDZ. The highest percentage of embryogenesis (93.33%) was observed in the presence of IAA at 0.05 mg/L and 0.5 mg/L Kin. Application of IBA at 0.05 mg/L with 0.5 mg/L Kin increased the embryogenic calli formation. Also, the highest regeneration percentage (83.33%) was found in all 4 treatments including 0.05 mg/L IBA, with 0.5 mg/L TDZ, 0.05 mg/L IBA with 0.5. 0 mg/L BAP, 0.05 mg/L 2,4-D with 1.5 mg/L BAP and 0.05 mg/L Pic, with 1.5 mg/L BAP. After transferring the explants to the second step culture media, the embryo-like structures (ELS) developed into plantlets. Finally, complete plantlets were obtained after rooting the plantlets on the MS medium with no supplement. In addition, the results of chromosome counting showed that among 10 regenerated plantlets, one plantlet was haploid, and 9 plantlets were diploid. | ||

| استفاده از گیاهان هاپلوئید در برنامههای بهنژادی به منظور تولید گیاهان کاملاً خالص در تعداد زیادی از گونههای زراعی میسر شده است. با توجه به پیشرفتهای اخیر در روشهای آزمایشگاهی، میتوان لاینهای خالص را در مدت زمان کوتاهی تولید کرد. در این مطالعه به منظور تولید گیاهان هاپلوئید از طریق کشت تخمدان، آزمایشی به صورت طرح کاملاً تصادفی در آزمایشگاه کشت بافت دانشگاه بینالمللی امام خمینی (ره) انجام شد. تیمارها شامل سطوح مختلف تنظیمکنندههای رشد گیاهی بودند. تخمدانها یک هفته پس از گلدهی برداشت و در محیط MS حاوی غلظتهای مختلف 0، 8/0 و 2/1 میلیگرم بر لیتر TDZ و BAP کشت داده شدند. بهترین کالوسها و درصد بالای کالوسزایی در محیط کشت حاوی 8/0 میلیگرم بر لیتر TDZ و BAP حاصل شد. سپس ریزنمونهها 3 هفته بعد به محیط تمایز شامل محیط MS تکمیل شده با 5 هورمون اکسین (NAA, IBA, IAA, 2,4-D, Pic) با غلظت 05/0 میلیگرم بر لیتر و 3 هورمون سیتوکینین (TDZ, BAP, Kin) با 3 غلظت متفاوت ( 5/0، 1 و 5/1 میلیگرم بر لیتر) در مجموع 15 تیمار هرکدام با 3 تکرار منتقل و هر دو هفته یکبار واکشت شدند. با توجه به نتایج مقایسه میانگین، بیشترین درصد کالوسزایی در حضور هورمون اکسین NAA با غلظت 05/0 میلیگرم بر لیتر و هورمون سیتوکینین TDZ با غلظت 5/1 میلیگرم بر لیتر حاصل شد. بیشترین درصد جنینزایی(33/93درصد) در حضور هورمونهای IAA با غلظت 05/0 میلیگرم بر لیتر و Kin با غلظت 5/0 میلیگرم بر لیتر مشاهده گردید. وجود هورمون IBA به میزان 05/0 میلیگرم بر لیتر همراه با Kin به میزان 5/0 میلیگرم بر لیتر سبب افزایش کالوسهای جنین داده شد. همچنین بیشترین درصد باززایی(33/83درصد) در 4 تیمار شامل 05/0 میلیگرم بر لیتر هورمون IBA با 5/0 میلیگرم بر لیتر هورمون TDZ، 05/0 میلیگرم بر لیتر هورمون IBA با 5/0 میلیگرم بر لیترBAP، 05/0 میلی-گرم بر لیتر هورمون 2,4-D با 5/1 میلیگرم بر لیتر هورمون BAP و 05/0 میلیگرم بر لیتر هورمون Pic با 5/1 میلی-گرم بر لیتر هورمون BAP حاصل گردید. بعد از انتقال ریزنمونهها به محیط کشت دوم ساختارهای جنین مانند به گیاهچه تبدیل شدند و در نهایت گیاهچههای کامل بعد از ریشه دار شدن روی محیط کشت MS بدون هورمون به دست آمد. علاوه بر این، نتایج شمارش کروموزوم نشان داد که در میان 10 گیاهچه باززا شده یک گیاهچه هاپلوئید و 9 گیاهچه دیگر دیپلوئید بودند. | ||
